چکیده

severe problem: when they are able to degrade a membrane structure, the structure of a membranebound enzyme may be altered simultaneously. This may result not simply in an inactivation of an enzyme (e.g. Mg2+-ATPase; Schoner et al., 1967; J0rgensen & Skou, 1969) but rather in an alteration of some, e.g. kinetic, properties. The present paper describes some results on the side effects ofEDTA and deoxycholate on the kinetic properties of Ca2+-ATPase (EC 3.6.1.3), from human erythrocyte membranes, produced when these compounds were used in the purification of this enzyme. The studies were performed to test the suitability of EDTA and deoxycholate as an aid in the purification procedure for Ca2+-ATPase of human erythrocyte membranes in comparison with a method published previously (Wolf, 1970, 1972).

Calcium ion-dependent p-nitrophenyl phosphate phosphatase activity and calcium ion-dependent adenosine triphosphatase activity from human erythrocyte membranes.

In the presence of ATP and of Mg(2+), human erythrocyte membranes show a phosphatase activity towards p-nitrophenyl phosphate which is activated by low concentrations of Ca(2+). The effect of Ca(2+) is strongly enhanced if either K(+) or Na(+) is also present. Activation of the p-nitrophenyl phosphate phosphatase by Ca(2+) reaches a half-maximum at about 8mum-Ca(2+) and is apparent only when th...

Human erythrocyte calmodulin. Further chemical characterization and the site of its interaction with the membrane.

Human erythrocyte and bovine brain calmodulins were indistinguishable by tryptic peptide mapping, indicating that the primary sequence of the two proteins is either very similar or identical. Calcium binding determinations of human erythrocyte calmodulin, by equilibrium dialysis and fluorescence titration, were in close agreement with previous studies on other calmodulins. The calcium-activated...

Calmodulin affinity chromatography yields a functional purified erythrocyte (Ca+ + Mg2+)-dependent adenosine triphosphatase.

The (Ca2+ + Mg2+)-dependent ATPase of human erythrocyte membranes was solubilized with deoxycholate and purified by calmodulin affinity chromatography to yield a functional enzyme. The method gave an enzyme purified 207-fold as compared with that of the erythrocyte membranes. The molecular weight of the ATPase was in the range 135 000-150 000, as revealed by a single major band after electropho...

The Effects of EGTA on the Quality of Fresh and Cryopreserved-Thawed Human Spermatozoa

Background: Sperm cryopreservation-thawing process has damaging effects on the structure and function of sperm, namely cryoinjury. Calcium overload has been reported as a postulated mechanism for sperm damage during the first steps after thawing. This study was designed to assess the intracellular calcium (Ca2+i) after cryopreservation and to clarify the role of a calcium chelator ethylene glyc...

Decreased ouabain-sensitive adenosine triphosphatase activity in the erythrocyte membrame of patients with chronic renal disease.

1. Ouabain-sensitive ATPase (adenosine triphosphatase) activity was measured in the erythrocyte membranes of twenty patients with chronic renal disease. Decreased activity was found in nineteen of the twenty patients. The average decrease was 38%. 2. In three patients erythrocyte sodium concentration exceeded 9.5 mmol/l of RBC and these patients had the most marked decreases in ouabain-sensitiv...
